摘要
目的建立一种从烟叶中提取分离茄尼醇的方法,以期为高纯度茄尼醇的生产提供可靠的技术指标。方法采用超临界CO2萃取法从烟叶中萃取得到茄尼醇粗品,通过硅胶柱色谱及结晶进行分离纯化。结果超临界CO2萃取茄尼醇的最佳实验条件为:萃取压力20MPa、萃取温度45℃、萃取时间3h。在此最佳实验条件下,茄尼醇的提取率为94.5%,纯度约为33.8%,并将此粗品经过硅胶柱色谱及后续结晶,得到纯度为93.4%的茄尼醇样品,折纯一次收率为70%。结论本方法简便可行,获得的茄尼醇纯度高、收率高。
OBJECTIVE To establish a way for extracting and separating solanesol from tobacco leaves. METHODS The extraction rate and the content of solanesol was determined by HPLC and the extraction rate and the content of solanesol were used as quality control criteria. The effect of different parameters such as extraction time, temperature and pressure on the supercritieal carbon dioxide extraction of solanesol was investigated. RESULTS The optimal extraction conditions were determined according to orthogonal experiments as entrainer with 95% ethanol (volume fraction), extraction pressure at 20 MPa, extraction temperature at 45 ℃ and extraction time for 3 h. The extraction rate of solanesol was 94.5% and the content of solanesol was 33.8 %.The content of purified sample was 93.4%. CONCLUSION This technology was feasible .The purity coefficient and yield of solanesol extracted by SFE and separated by column chromatograph were high.
