不同浓度健脾解毒中药对肝癌裸鼠的抗癌作用

Anticancer activities of Jianpi Jiedu Decoction on athymic mice with hepatocellular carcinoma

[目的]探讨不同浓度健脾解毒中药对间接原位移植肝癌裸鼠模型的抗癌作用及最佳药效浓度。[方法]采用人肝细胞癌细胞株Bel-7402间接原位移植，造雄性肝癌裸鼠模型90只，造模后24h随机分9组，即不同浓度健脾解毒复方中药A、B、C、D、E、F、G组，替加氟207化疗（FT）组，0．85％氯化钠（NS）组，8周后处死裸鼠，观察各组荷瘤裸鼠的生存时间、体重变化、肿瘤形成情况、肝脾指数等。另外10只雄性裸鼠不造模作为正常对照（DZ）组。[结果]造模成功率为76．14％，实验结束获得有效病理标本67例，成瘤以多发小结节为主；各中药组的生存时间和生存率均优于FT组（P〈0．05），中药D、E组的生存时间〉NS组（P〈0．05）；各中药组荷瘤裸鼠治疗后体重有增加（P〈0．05），FT组治疗后体重明显下降（P〈0．05）；中药组的肝指数和脾指数均〉FT组（P〈0．05）。[结论]健脾解毒复方中药可以增加荷瘤裸鼠的体重，改善营养状态，延长带瘤生存时间。健脾解毒复方中药抗癌作用的有效范围为7．5～20．0g／kg。

[Objective]To research the effect and optimization drug concentration of Jianpi Jiedu Decoction （JPJDT） in treating experemental HCC transplanted with Bel-7402. [Methods] Ninety male BALB/c athymic mice models were indirect orthotopic transplanted with Bel-7402. After 24 h of postoperation, the 90 athymic mice were distributed randomly into JPJDT groups： A, B, C, D, E, F, G, NS, FT each group had 10 athymic mice. Group A to G were treated by intragastric administration with JPJDT that had been deliquated into 7 kinds of concentration for 8 wk. Group NS were treated by intragastric administration with Sodium Chloride for 8 wk. Group FT were were treated by intragastric administration with FT207 （Tegafur） for 8 wk. Live time, change of body weight, tumorigeness, exponent of liver and spleen were observed. Ten male BALB/c athymic mice treated NS were as normal control （DZ）. [Results]1. The dissection confirmed that the achievement ratio of making model was 67/88, most tumor of 67suitable patho-samples were multi-nodule. 2. Survival state： The live time and survival rate of Group JPJDD were much longer than Group FT（P〈0. 05）, the live time in Group D and E was much longer than Group NS（P〈0. 05）. 3. Body weight state： The body weight of Group JPJDD was increased after treatment （P〈0. 05）, while The body weight of Group FT was decrease obviously after treatment （P〈0. 05）. 4. Dissection state： The index of liver and spleen in Group JPJDD was higher than Group FT （P〈0. 05）, the index of spleen in Group C, E, F was higher than Group FT （P〈0. 05）, meanwhile The index of spleen of Group C higher than Group D （P〈 0. 05）. [Conclusion]JPJDD can extend the live time, increase survival rate and increase the body weight of athymic mice with hepatocellular carcinoma. The optimization drug concentration of JPJDT is from 7. 5 g/kg to 20. 0 g/kg.
