摘要
采用四甲基偶氮唑盐(MTT)观察3,4,5-三羟基苯甲酸(TBA)对人肝癌SMMC-7721细胞的增殖抑制作用;通过流式细胞仪检测细胞凋亡、细胞内活性氧(ROS)及线粒体膜电位的变化,用比色法测定Caspase-9和Caspase-3蛋白活性,探讨TBA诱导SMMC-7721细胞凋亡的分子机制.研究结果表明,TBA对SMMC-7721细胞生长具有显著的抑制作用并诱导其凋亡,诱导SMMC-7721细胞凋亡作用可能通过线粒体信号传导通路实现.
Trihydroxybenzoic acid show the anti-tumor potency from many previous investigations, but the research for the inhibitory effect and mechanism on SMMC-7721 cells has not been regarded so far. MTT assay was used to evaluate the anti-proliferation effect of trihydroxybenzoic acid on SMMC-7721 cells and human PBMC. ROS production, apoptotic cells and the mitochondrial membrane potential were detected by flow eytometry. The activities of Caspase-3,9 were measured by colorimetric method. The results show that trihydroxybenzoic acid significantly inhibit the proliferation of SMMC-7721 cells in dose and time-dependent pathways. Compared with control group, trihydroxybenzoic acid caused the increase of ROS levels and a concomitant dissipation of the mitochondrial membrane potential, activities caspase-9 and caspase-3, triggering apoptosis of SMMC-7721 cells. Mitochondrial-dependent pathways could involved in trihydroxybenzoie acid induced SMM-7721 cells apoptosis.
