摘要 目的建立防风药材中4种有效成分含量同时测定的方法,以比较不同产地防风药材及不同部位中有效成分的含量,为防风药材质量标准的制定及防风的资源开发提供科学依据。方法采用RP-HPLC法。色谱柱:Diamonsil C18柱(200mm×4.6mm,5μm);流动相:甲醇-水,梯度洗脱;流速:1.0mL·min^-1;检测波长:254mm;柱温:30℃。结果升麻素、升麻素苷、5-O-甲基维斯阿米醇苷、亥茅酚苷能够达到基线分离;升麻素苷和5-O-甲基维斯阿米醇苷质量浓度在1.92~400mg·L^-1内与峰面积呈良好的线性关系(r=0.9999、0.9998,n=5),平均加样回收率分别为97.5%、97.7%,RSD分别为1.3%、2.8%(n=9);升麻素、亥茅酚苷质量浓度在0.96~200mg·L^-1内与峰面积呈良好的线性关系(r=0.9999、0.9999,n=5),平均加样回收率分别为99.3%、97.7%,RSD分别为2.0%、1.7%。结论本方法为防风药材的资源开发提供了参考,可用于防风药材的质量控制。 Objective A high performance liquid chromatographic method was established for analysis of four components from different habitats and different parts of Saposhnikovia divaricata (Turcz.) Schischk. Methods Diamonsil C18 column( 200 mm × 4.6 mm ,5 μm)was used with a mixture of methanol and water as the mobile phase, gradient elution was performed. The flow rate was 1.0 mL. min^-1, detection wavelength was set at 254 nm, column temperature was set at 30℃. Results The linear range of prim-O-glucosylcimifugin,4'-O-β-D-glucosyl-5-O-methylvisamminol and cimifugin, sec-O-glucosylhamaudol were 1.92 - 400 mg. L^-1 and 0.96-200 mg.L^-1, respectively. The average recoveries of prim-O-glucosylcimifugin and 4'-O-β-D- Glucosyl-5-O-methy-lvisamminol were 97.5% and 97.7% with RSD of 1.3% and 2.8% ,respectively. The average recoveries of cimifugin and sec-O-glucosylhamaudol were 99.3% with RSD of 2.0% ,97.7% with RSD of 1.7 %, respectively. Conclusions The method is simple, accurate and sensitive with good reproducibility. It can provide references for quality control and resources development of the medicinal materials.
