摘要 细胞周期蛋白依赖性激酶(Cdks)是细胞周期调节的核心分子,其与细胞周期蛋白(Cyclins)、细胞周期蛋白依赖性激酶抑制因子(CKIs)等组成细胞周期调控网络系统。细胞周期调控机制的异常与肿瘤的发生发展密切相关,Cdks和Cyclins的异常表达、CKIs的缺失以及检验点的异常都将使细胞周期紊乱,细胞增殖失控,最终发生癌变。Cdks与Cyclins结合形成的复合物对细胞周期进程具有正调节作用,其高表达和激酶活性异常可促进卵巢癌的发生、发展。就Cdks的生物学功能、活性调节及其在卵巢癌发生、发展中的作用进行综述。 In spite of the clinical importance of epithelial ovarian cancer (EOC), little is known about the pathobiology of its precursor lesions and pmgession of the disease. Cyclin-dependent kinases (cdks), composed of the cdks, cyclins and cyclin-dependent kinase inhibitors (CKI's), play critical roles in the regulation of the cell-cycle. Alterations in these molecules can result in uncontrolled cell proliferation, which is a distinctive feature of human cancers. Cdk-cyclin interactions positively control cell-cycle progression, with overexpression or high kinase activity of these molecules leading to unregulated cell-cycle progression and uncontrolled cell proliferation. This review describes the current state of knowledge about the alterations of cdks in the genesis and progression of EOC.
