大鼠创伤性休克早期血液单个核细胞释放HSP70的变化及甘氨酸的影响 被引量:1

摘要 目的研究观察血液单个核细胞(blood mononuclear cells,BMCs)释放HSP70、TNF-α和IL-6水平的动态变化及甘氨酸的干预影响。方法建立创伤性休克动物模型,88只Wistar大鼠随机分成3组:治疗组(T组)、休克组(S组)和对照组(C组)。在复苏开始时,T组大鼠经颈静脉输入甘氨酸液0.5mL(甘氨酸浓度为100mg/kg的生理盐水);S组输以相同体积的生理盐水;C组仅行腹腔麻醉。分别于复苏后3、6、12和24h共4个时相点活杀大鼠。采用酶联免疫吸附法(ELISA)检测BMCs释放HSP70、TNF-α和IL-6的水平。结果S组大鼠复苏后3h BMCs释放HSP70、TNF-α和IL-6水平均出现升高,复苏后6h达高峰。与S组相比,T组大鼠各个时间点BMCs释放HSP70水平均明显增高,而释放TNF-α和IL-6水平均明显降低(均P<0.01)。结论甘氨酸可促进BMCs释放HPS70、抑制BMCs释放TNF-α、IL-6,可望起到防治创伤性休克后SIRS及MODS的作用。 [Objective] To investigate the effects of glycine on the release of HSP70, TNF-α and IL-6 from blood mononuclear cells (BMCs) in rats with traumatic shock. [Methods] The traumatic shock model was established and 88 Wistar rats were divided randomly into three groups: treatment group (T group), shock group (S group) and control group (C group). At the beginning of resuscitation, the rats in T group received 0.5 mL normal saline contained 100 mg/kg glycine by external jugular vein, those in the shock group received only 0.5 mL normal saline. And the rats in the C group received only intrapefitoneal anesthesia. The rats in the three groups were killed at 3, 6, 12 and 24 h after resuscitation respectively. The levels of HSP70, TNF-α and IL-6 released from BMCs were detected by ELISA. [Result] The levels of of HSP70, TNF-α and IL-6 released from BMCs of rats in S group increased at 3 h and reached the peak value at 6 h after resuscitation. Compared with S group, the level of HSP70 in BMCs in T group increased significantly and the levels of TNF-α and IL-6 decreased significantly (all P 〈0.01). [Conclusion] Glycine can promote the release of HSP70 and reduce the release of TNF-α and IL-6 from BMCs after traumatic shock. It may play an important role in the prevention and treatment of the secondary SIRS and MODS after traumatic shock.
