摘要
目的探讨干扰素诱导蛋白10(IP-10)对体外培养变应性接触性皮炎(ACD)小鼠T淋巴细胞亚群的影响,探讨ACD的发病机制。方法建立ACD小鼠模型,分离纯化小鼠脾淋巴细胞,并和不同浓度的IP-10共同培养,应用流式细胞术检测与IP-10共同孵育的小鼠脾淋巴细胞中CD4+、CD8+和CD3+水平,以方差分析进行结果分析。结果IP-10各浓度组体外培养的ACD小鼠CD4+、CD8+、CD3+T淋巴细胞的含量均高于对照组,但仅高浓度组与对照组之间的差异具有统计学意义(P<0.01)。结论IP-10可升高成熟T淋巴细胞比率,非特异性地增加Ⅳ型变态反应主要效应细胞CD4+T及CD8+T亚群的表达,扩大变应性接触性皮炎的免疫反应。
Objective To study the function of interferon-inducible protein-10 (IP-10) in allergic contact dermatitis(ACD) through observing its effect on T-lymphocytes. Methods Establish the ACD mouse model with 2, 4-Dinitrochlorobenzene (DNCB). After the separation and purification of spleen lymphocytes from mice, spleen lymphocyte was co-cuhured with interferon protein (IP)-10. Flow cytometry (FCM) was used to detect the ratio of CD4^+, CD8^+ and CD3^+. ANOVA method was used to analyze the difference on CD4^+, CD8^+ and CD3^+ among the four groups. Results The ratio of CD4^+, CD8^+ and CD3^+ express on the ACD mouse model T-lymphocyte in vitro in the three groups was much higher than that in the control group. The expression in the 100ng/ml groups was significantly higher than that in the contol group (P〈0.01). Conclusion IP-10 could increase the ratio of T-cell subset, including CD3^+, CD4^+ and CD8^+, which promoting the development of ACD.
