苯丁酸钠对体外培养人宫颈癌Hela细胞增殖抑制作用 被引量:3

摘要 目的:探讨苯丁酸钠(SPB)在体外诱导人宫颈癌Hela细胞生长抑制、分化和细胞周期阻滞。方法:培养Hela细胞,应用MTT比色法观察苯丁酸钠对Hela细胞的生长抑制作用,倒置相差显微镜观察细胞形态改变,流式细胞术分析细胞周期。结果:苯丁酸钠可明显抑制Hela细胞的生长,呈时间一剂量依赖性,并出现典型的细胞形态学改变;苯丁酸钠诱导Hela细胞阻滞于G0/G1期,S期细胞数减少。结论:苯丁酸钠在体外抑制人宫颈癌Hela细胞的增殖,使阻滞于G0/G1期。这可能是苯丁酸钠抗宫颈癌作用的重要机制之一。 Objective : To investigate the effects of sodium phenylbutyrate on the proliferation, differentiation,phasegrowth arrest in Human cervical cancer cell line HeLa in vitro. Methods : HeLa cell was treated with SPB at different concentrations. MTT assay was adopted to describe the proliferation of HeLa cell. interred microscope was used to find out morphological changes in HeLa cell. Cell cycle was determined by flow cytometry. Results : sodium phenylbutyrate inhibited the proliferation of HeLa cells significantly in time and dose dependent manner. The characteristic morphological changes in Hela cell were observed by interred microscope, sodium phenylbutyrate induced cell cycle arrest at G0/G1 phase and cell number in S phase was decreased. Conclusion: It was demonstrated that sodium phenylbutyrate may inhibit the proliferation of Hela cell in vitro and cause cell cycle arrest at G0/G1 phase. This may be one of the important mechanism of inhibition induced by sodium phenylbutyrate on Human cervical cancer.
