应用SELDI技术检测精浆抗精子抗体阳性不育患者精浆中差异蛋白

Screening of seminal plasma biomarkers in infertile patients with positive antisperm antibody in seminal plasma by SELDI technique

摘要 目的探讨用蛋白质芯片技术筛选精浆抗精子抗体阳性不育患者精浆中蛋白质表达谱,寻找精浆中的差异蛋白。方法采用表面增强激光解吸电离飞行时间质谱技术(SELDI-TOF-MS)和CM10蛋白质芯片对30例精浆抗精子抗体阳性不育患者精浆和30例健康人精浆的蛋白质谱图进行了检测,使用PBSⅡ-C型蛋白质芯片阅读机读取数据,获得的结果采用CIPHERGEN公司的Biomarker wizard和Biomarker Patterns System软件分析。结果精浆抗精子抗体阳性不育患者与正常人精浆蛋白质谱相比有12个显著差异蛋白质,其中6个蛋白质在患者精浆中高表达,6个蛋白质在患者精浆中低表达。Biomarker Patterns System软件用12个显著差异蛋白质中的2个标志分子建立精浆抗精子抗体阳性不育患者诊断的分类树模型。检测灵敏度为90%、特异性为93.3%。结论实验证明利用蛋白质组学和生物信息学方法可以从精浆中筛选出精浆抗精子抗体阳性不育患者相关的标志蛋白质,而且蛋白质芯片技术对于发现和筛选精浆中的精浆抗精子抗体阳性不育患者的标志蛋白质是一种有效、快速的工具。 Objective: To screen out semen protein profiling of infertile patients with positive antisperm antibody in seminal plasma by surface - enhanced laser desorption / ionization time of flight mass spectrometry ( SELDI - TOF - MS) for discovering the discriminatory proteins. Methods: Proteomic spectra were generated by mass spectrometry in 60 cases, including 30 cases of infertile patients with positive antisperm antibody in seminal plasma that had been pathologically confirmed with 25 to 34, and 30 cases of healthy control aged 26 -35. 80 spectra obtained were used to train and develop a decision tree classification algorithm. Results: A total of 12 distinguished proteomic peaks were detected, 2 peaks were used to build a proteomic pattern. The results yielded a sensitivity of 90%, specificity of 93.3%. Conclusion: SELDI -TOF -MS offers a unique platform for the proteomic detection of infertile patients with positive antisperm antibody in seminal plasma. It also offers a noninvasive method to further study the proteomic changes in the development and progression of infertile patients with positive antisperm antibody in seminal plasma.
