摘要
目的:观察抑癌基因ASPP家族在不同肿瘤细胞株中的表达水平,探讨其在肿瘤的发生、发展及预后中的作用。方法:应用单核细胞分离液分离正常人血液单核细胞,用DMEM培养液培养细胞株jurkat、k-562、HL-60、成纤维细胞、HT-29、所得细胞用Tripure分离试剂提取总RNA。最后用RT-PCR检测ASPPmRNA的表达水平。结果:ASPP1与ASPP2mRNA在正常人细胞中的表达水平比肿瘤细胞株的表达水平高(P<0.05)。iASPP的表达量除在HT-29细胞株中与正常细胞差异无统计学意义(P>0.05),其他肿瘤细胞株的表达量均高于正常细胞的表达量(P<0.01)。结论:抑癌基因ASPP家族的促p53细胞凋亡功能在正常细胞转变为恶性的过程中有重要的作用。
Objective:To observe ASPP family gene expression levels in tumor cell lines in order to study their effects on tumor occurrence, development and prognosis. Method:Normal blood monocyte was isolated using monocyte isolation reagent. Cell lines of jurkat, k-562, HL-60, fibrobtast, HT-29, HCTII6, MCF-7, A549 and HEPG-2 were cultured using DMEM culture medium. RNA was extracted from these cultured cells using tripure TM isolation reagent and ASPP gene family expression level was detected by RT PCR assay. Result: ASPP1 and ASPP2 mRNA expression levels were higher in normal cell than those in tumor cell lines(P〈0. 05). iASPP mRNA expression level was higher in normal cell than that in tumor cell lines(P〈0. 05), except A549 and HT-29 (P〉0.05). Conclusion:The apoptotic function of ASPP family on p53 is important in the transformation course of normal cell to malignant cell.
