摘要
目的:建立HPLC-ELSD同时测定动物类药材牛胆粉中胆酸、去氧胆酸和牛黄胆酸钠3种成分含量的方法。方法:色谱柱为Agilent Zorbax SB-C18(4.6mm×150mm,5μm),流动相采用乙腈-0.2%甲酸进行梯度洗脱,流速1mL.min-1,采用蒸发光散射检测器检测。结果:胆酸、去氧胆酸和牛黄胆酸钠的线性范围分别为0.45～4.48,0.21～4.18,0.23～4.56μg;加样回收率(n=9)分别为96.39%,96.78%,97.37%。结论:本方法简便、灵敏、准确,可用于牛胆粉药材中胆酸、去氧胆酸和牛黄胆酸钠3种成分的含量测定。
Objective ：To develop an HPLC - ELSD method for simultaneous determination of cholic acid,deoxycholic acid and sodium taurocholate in Niudan powder（ from Bos Taurus domesticus Gemelin）. Methods：An Agilent Zorbax SB - C18 column （4. 6 mm × 150 mm,5 μm）was applied ;a gradient elution solvent system composing of acetonitrilc and 0. 2% formic acid was used as the mobile phase,and the flow rate was 1.0 mL · min^-1. An ELSD was applied as detector. Results：The results were obtained that quantitative linear ranges of three components were 0. 45 -4. 48,0. 21 -4. 18,0. 23 -4. 56 μg respectively,and the average recoveries（n =9）were 96. 39% ,96. 78% ,and 97. 37% respectively. Conclusion ：A specific, sensitive, simple and reproducible HPLC -ELSD method has been established for simultaneous determination of the three cholic acid derivatives in Niudan powder.
