RP-HPLC法测定藏药细果角茴香中原阿片碱的含量

RP-HPLC determination of protopine in Tibet medicine Hypecoum leptocarpum Hook. f. et Thoms

目的:建立一种藏药细果角茴香中原阿片碱含量的HPLC测定方法。方法:采用Thermo C18柱(4.6mm×250mm,5μm),以甲醇-0.3%三乙胺(64:36)为流动相,流速1.0mL.min-1,检测波长285nm。结果:原阿片碱进样量在0.24～1.20μg范围内呈现良好的线性关系(r=0.9999),平均加样回收率(n=5)为99.98%(RSD=1.6%),2个地点的药材中原阿片碱的含量分别达到2.37%,0.10%。结论:此法便捷、准确,重复性好,为藏药细果角茴香的研究及其质量控制提供了一定的参考。

Objective：To establish an RP - HPLC method to determine the content of protopine in Hypecoum leptocarpum Hook. f. et Thoms.. Method ：The analysis was performed on a Thermo C18 （4. 6 mm × 250 mm,5 μm） eluting with a mixture of methanol and 0. 3% triethylamine（64： 36） as mobile phase with UV detection at 285 nm. Result： The linear range of protopine was 0. 24 - 1.20 μg（ r = 0. 9999）, and the average recovery （ n = 5 ） was 99. 98% （ RSD = 1.6% ）. The contents of two samples were up to 2. 37% ,0. 10%, respectively. Conclusion：This method is accurate and repeatable, and can be used for the quality control of Hypecoum leptocarpum Hook. f. et Thorns..
