摘要
小鼠体外成熟卵母细胞组的乙醇激活率显著低于体内成熟卵母细胞组水平。在体外成熟过程中[培养液:TCM-199+FCS(20%)+hCG(2IU/ml)],卵丘细胞的存在极显著地降低了成熟卵的乙醇激活率,卵激活率由58.7%(裸卵)降为13.8%(卵丘卵母细胞复合体)。超排卵子随着卵龄的增加,卵子的乙醇激活率逐渐升高,而且体内老化卵的激活率高于体外老化同龄组的水平。
The ability of mouse oocytes to undergo parthenogenetic activation using ethanol was investigated Comparing with postovulatory oocytes aged in the oviduct,mouse oocytes achieved only a low level of activation competence when they were retrieved from follicles at the germinal vesicle stage and matured in vitro.The maximal activation rate of postovulatory oocytes was over 80%,and only a maximum of 58 7% was obtained in oocytes matured in vitro.When oocytes were matured in TCM 199+FCS(20%)+hCG(2 IU/ml),the proportion of activated oocytes were lower( P< 0 01)in those matured with cumulus cells (13 8%) than in those matured without them (58 7%).The activation rate of oocytes matured in vitro was influenced by the maturation time.Oocytes matured in vitro for 12h,18h,24h,the activation rates were 5 3%,34 3%,58 7% in denude oocytes ,and 0,3 7%,13 8% in cumulus oocyte complexes respectively.The activation rate of oocytes improved while postovulatory oocytes were aged in vivo or in vitro and the maximum percentages of activated embryos was higher in eggs aged in vivo than that in eggs aged in vitro.
