利用七叶灵显色技术检验和判断β-葡萄糖苷酶的研究 被引量:18

摘要 七叶灵(6,7-2-羟基-香豆素-β-D-葡萄糖苷)在β-葡萄糖苷酶的作用下能生成葡萄糖和七叶苷原(6,7-2-羟基-香豆素),七叶苷原能和Fe3+作用呈现棕黑色。七叶灵显色技术用于β-葡萄糖苷酶高产菌株的筛选、β-葡萄糖苷酶提纯时的样品检测、非变性聚丙烯酰胺凝胶活性染色的应用结果表明,以七叶灵为作用底物检验和判断β-葡萄糖苷酶的新方法具有高分辨率和灵敏性,而且操作简易、快速、经济。 Based on the well-established reaction of β-glucosidase with eseulin (6,7- dihydroxycoumarin - β-glucoside), Esculin were used as a substrate for Screening of the strain producing β-glucosidase from a large number of organisms, detection β-glucosidase from Chromatographic fractions and identification β-glucosidase in polyacrylamide gels. The experiment results showed that it was a novel technique for the detection and identification of β-glucosidase with high resolution and sensitivity and that can be performed easily, rapidly, and at a minimal cost.
