摘要
本研究以马铃薯栽培品种鲁马一号为实验材料,应用正交试验设计方法,以最少量的典型试验资料研究了再生系统主要因子对马铃薯外植体(叶园片)再生率的影响,并且利用改进的单纯形法对再生系统进行优化,得到了良好的组织培养条件和优化的再生系统。试验结果表明:此方法能大大缩短优化再生条件的筛选周期,可靠性高。
Potato cultivar Luma-1 was used to study the effect of multifactors on plant regeneration frequency.The method used was a combination of orthogonal test and improved simplex algorithm.The optimal factors for potato tissue culture as determined by using this method were:AgNO 3 4mg/L,pH 5.8, ABA 0.2mg/L,4 weeks of seedling age,14 days of dark treatment period.Our results show that this method is very reliable,and could greatly shorten the screening period of optimizing regeneration conditions.This method could also be used for factor optimization of tissue culture system for other plants.
