麻疹病毒L4株基因组全序列的测定

摘要 将麻疹病毒L4株毒种接种于CEC细胞,传代培养及鉴定合格后,用Trizol法提取总RNA,进行分段RT-PCR扩增,并将PCR产物精制后测序分析。结果表明,成功测出麻疹病毒L4株15894 nt全基因组序列,为了解其基因组结构与生物功能的关系及研究开发麻疹病毒新型疫苗而奠定基础。 Measles virus strain Leningrad 4 (L4) was transfected into CEC cells, and after culture passaging for several generations and qualified, its total RNA were extracted with Trizol method and carried out RT-PCR amplification fragment by fragment, then analyzed its sequence after refinement. The results showed that the whole sequence of 15894 nt RNA of measles virus strain L4 was determined successfully, this had established foundation to study the relation between genomic structure and biological functions and develop new type measles virus vaccine.
