摘要
以西北农林科技大学林学院苗圃4年生叉子圆柏茎段为外殖体建立无菌苗,探讨了不同培养基和不同生长调节物质对外植体分化、增殖和生根的影响,并对栽培基质与移栽技术进行研究。结果表明适宜的诱导培养基为1/2MS+6-BA5+NAA0.3;继代培养基为1/2MS+6-BA5+NAA0.3,增殖率为4.0;生根培养基为1/2WPM+6-BA0.5+NAA1+0.2%活性炭;移栽基质为蛭石+珍珠岩+沙(1:1:1)效果最好,成活率93%。
Cut stems of four years' Sabina Vulgaris were used as explants to establish seedlings. Influence of different growth regulating substances and media on explants differentiation, multiplication and rooting were studied. Cultivation techniques as well as cultivation media used were described. The results showed that the best medium for inducement was 1/2MS+ 6-BA5 +NAA0. 3; the best media for the multiplication was 1/2MS + 6-BA5 +NAA 0.3; the multiplication rate was 4.0; the best medium for rooting was 1/2WPM+6--BA0.5+NAA1+0.2% active carbon; and that best medium for transplanting was vermiculite+pearlite+sand with a ratio of 1:1:1, making the survival rate 93%.
