原花青素诱导前列腺癌PC-3细胞凋亡的机制研究 被引量:7

摘要 目的:探讨原花青素在体外诱导人前列腺癌PC-3细胞凋亡的机制。方法:体外培养人前列腺癌PC-3细胞株,用100,200,300μg/ml原花青素作用24 h。荧光素标记的连接素Ⅴ(An-nexinⅤ-fluoresein isothiocyanate,AnnexinⅤ-FITC)和碘化吡啶(propidium iodide,PI)双染检测原花青素对PC-3细胞凋亡的影响,采用流式细胞术检测以不同浓度原花青素作用后PC-3细胞线粒体膜电位(Δψm)的变化。结果:100μg/ml原花青素作用细胞24 h,5.64%的细胞出现早期凋亡,84.7%的PC-3细胞呈现线粒体膜电位降低;300μg/ml原花青素组PC-3细胞凋亡率和坏死率分别为44.86%、50.81%。结论:原花青素可在体外诱导PC-3细胞凋亡,其机制与降低线粒体膜电位有关。 Objective: To observe the effect of procyanidins on apoptosis in cultured prostate carcinoma PC-3 cells and its correlation with mitochondrial membrane potential (△ψm) changes. Methods. After treated with procyanidins 100; 200, 300 μg/ml respectively, PC-3 cells were doubly stained by Annexin V-fluoresein isothiocyanate (FITC) and propidium iodide (PI). The procyanidins-induced apoptosis and necrosis were detected by flow cytometry(FCM). The change of △ψm was analyzed with FCM after Rhodamine 123 staining Results: After treated with procyanidins 100μg/ml for 24 hours, the apoptosis rate of PC-3 cells was 5.64%, and △ψm was significantly decreased in 84.7% of PC-3 cells. The apoptosis rate and necrosis rate of PC-3 cells was 44.86 %, 50.81% in the group treated with procyanidins 300μg/ml, respectively. Conclusion. Procyanidins could induce apoptosis and necrosis in PC-3 cells, which might relate with down-regulation of mitochondrial △ψμm.
