摘要
目的采用HPLC法和uV法测定川渝产山银花中的绿原酸和总绿原酸。方法色谱柱为DikmaKromasil C18柱(250mm×4.6mm,5μm);流动相为乙腈-0.4%磷酸溶液(12:88);流速0.8ml·min-1;检测波长326nm;柱温30℃;进样量10μl。结果绿原酸0.8~8.0μg具有良好的线性关系(r=0.9998),平均回收率为98.75%,RSD=0.29%。结论方法操作简便、快速、具有良好的重复性和回收率,可作为川渝产山银花中绿原酸和总绿原酸的含量测定方法。
OBJECTIVE To establish the method for content determination of chlorogenic acid and total chlorogenic acid in Flos Lonicerae from Sichuan and Chongqing. METHODS The content of chlorogenic acid was detected by HPLC, and total chlorogenic acid was detected by UV. The separation was carried out on a Dikma Kromasil C18 column (250 mm×4. 6mm,5 μm). The mobile phase was acetonitrile -0. 4% phosphoric acid. The flow rate was 0. 8 ml. min-1. The detective wavelength was set at 326 nm,The col- umn temperature was 30℃ and the sample volumn was 10 μl. RESULTS The calibration curve showed good linearity over the range of 0. 8 - 8.0 μg( r = 0. 9998). The average recovery was 98. 75% with RSD of 0. 29%. CONCLUSION This method is simple, rapid, efficient, sensitive, accurate and with good repeatability and recovery, it can be used as a quantitative analysis method for this preparation.
