两相系统中生物转化茴脑生成茴香醛 被引量:7

摘要 考察了假单胞菌BT-13在有机溶剂-水两相系统中生物转化茴脑生成茴香醛的过程,初步探讨了有机溶剂极性和加入量、培养基配方、转化时间、转化温度、摇床转速、装液量及pH等条件对游离细胞转化茴脑合成茴香醛的影响。结果表明:适宜有机溶剂为乙酸乙酯,加入量控制在体积分数10%为宜;适宜的培养基为改良马丁氏培养基,培养方式采用摇床培养。在装液量为20 mL每150 mL三角瓶,培养基初始pH控制在6.5,温度30℃,转速150 r/m in下转化30 h,茴香醛的摩尔转化率可达7.7%。在乙酸乙酯-水两相系统中,用海藻酸钙固定化细胞进行转化,茴香醛的摩尔转化率有所提高,达12.6%。 The biotransformation process of anethole to anisaldehyde by Pseudomonas sp. BT - 13 was carried out in aqueous-organic solvent biphasic systems. Effects of some key factors such as organic solvent polarity, relative amount of the organic solvent, the component of culture medium, conversion time,temperature, amount of medium with repect to the capacity of the container, rotating speed of the rotary shaker and medium initial pH on the biotransformation of anethole to anisaldehyde by free cell were investigated. Results showed that the suitable organic solvent was ethyl acetate, its relative amount was 10% ,the suitable medium was the modified Martin's medium, amount of medium with respect to the capacity of the container was 20 mL in a 150 mL flask, medium initial pH was 6. 5, temperature was 30 ℃ ,rotating speed of rotary shaker was 150 r·min-1 ,and conversion time was 30 h. Under optimun conditions with calcium alginate immobilized cells, the molar tranformation ratio of anisaldehyde was increased to 12.6% as compared to 7.7% transformation with free cells.
