高浓度CO2气调对药材甲羧酸酯酶活性的影响 被引量:3

Effect of CO_2 Enriched Atmospheres on Activity and Kinetic Index of Carboxylesterases from Adults of Stegobium paniceum(L.)

摘要 用高浓度CO2气体胁迫处理药材甲Stegobium paniceum(L.)成虫及离体羧酸酯酶,通过终点法测定酶活性及其动力学参数变化,分析气调胁迫对羧酸酯酶性质的影响.结果显示,处理6,9,12 h,羧酸酯酶比活力分别为0.412 mmol/(mg.min)、0.439 mmol/(mg.min)、0.418 mmol/(mg.min),与对照0.358 mmol/(mg.min)相比,分别升高了14.96%,22.58%,16.69%,差异均达到显著水平.处理时间延长至15 h,比活力降低了2.73%.羧酸酯酶酶液经持续通入高浓度CO2于37℃水液中离体处理10 min,比活力显著降低.离体处理亦导致羧酸酯酶动力学参数发生相应变化. On stress of carbon dioxide (CO2) enriched atmospheres, the timeliness response activity of carboxylesterases (CarE) from adults of Stegobium paniceum (L.) and its kinetic index were investigated by endpoint method colorimetry. Results showed that, S. paniceum(L. ) were exposed to the CA 6,9 and 12 h, the activity of Care from the insects was 0. 412 mmol·mg^-1 pro · min^-1,0. 439 mmol · mg^-1pro · min^-1 and 0.418 mmol · mg^-1 pro · min^-1 respectively which increased 14.96% ,22.58% and 16.69% compared to the control. However, the activity declined 2.73% when the insects were exposed to the CA for 15 h. The unorganized ferment CarE was treated with CO2 enriched atmosphere for 10 min at 37 ℃, its activity was declined 31.23 %. Moreover, the stress on unorganized ferment also contributes to the change of kinetic index.
