摘要
目的探讨何首乌饮对亚急性衰老大鼠卵巢细胞凋亡和相关基因Fas和FasL及其蛋白表达的影响。方法采用D-半乳糖连续腹腔注射致亚急性衰老大鼠模型,同时以何首乌饮灌胃作为延缓衰老组;造模后应用脱氧核糖核苷酸末端转移酶介导的缺口末端标记(TUNEL)法检测各组大鼠卵巢细胞凋亡情况;测定各组Fas和FasL的信使核糖核酸(mRNA)水平;SP免疫组化法检测卵巢细胞Fas和FasL蛋白的表达情况。结果与正常对照组比较,模型组大鼠卵巢细胞中有凋亡细胞数增多现象、Fas和FasL的mRNA和蛋白表达均显著增强(P<0.01);延缓衰老组较模型组凋亡率明显下降(P<0.01),并抑制Fas和FasL的表达,呈现量效关系,以何首乌饮高剂量组效果作用最佳。结论何首乌饮方剂能延缓衰老大鼠卵巢细胞凋亡的发生,其作用机制可能是通过干预Fas/FasL的表达,阻止死亡信号的转导,抑制卵巢细胞凋亡。
Objective To investigate the effects of Heshouwuyin (HSWY) on apoptosis and the expressions of Fas and Fasl protein in ovary of aging rats. Methods The model of sub-acutely aging rats were made by injecting D-gal into the rats, abdominal cavity continually. After lavaging HSWY, the apoptosis rate and the levels of Fas and FasL mRNA were measured by TUNEL and RT-PCR; the expressions of Fas/Fasl protein were detected by SP immunocytochemistry. Results Compared with normal control, the number of apoptosis cells and the expressions of Fas/Fasl mRNA, protein in model group were significantly higher (P 〈 0. 01 ). The apoptosis rate and the expressions of Fas/Fasl mRNA, protein in HSWY group were significantly decreased( P 〈 0. 01 ), and those in high dose group were the lowest. Conclusions HSWY can delay the incidence of ovary cell apoptosis. By interfering the expressions of Fas/Fasl, HSWY may prevent transmission of death signal and inhibit ovary cell apoptosis.
