宫内缺氧对子代大鼠老年期脑白质GFAP表达的影响 被引量:1

摘要 目的研究宫内缺氧(intrauterine hypoxia,IH)对子代大鼠老年期脑白质胶质纤维酸性蛋白(glialfibrillary acidic protein,GFAP)表达的影响。方法建立SD大鼠宫内缺氧模型,成年后暴露于高脂饮食(high-fat diet,HF)和(或)追加缺氧(second hypoxia,SH),免疫组织化学检测子代大鼠老年期脑室旁白质GFAP表达并行图像分析。结果宫内缺氧的主效应可使子代大鼠老年期脑白质的GFAP表达增加(P<0.05),其中IH与HF、IH与SH、HF与SH间存在一阶交互作用(均P<0.05),IH、HF与SH3因素间存在二阶交互作用(P<0.05)。结论宫内缺氧可使子代大鼠老年期脑白质GFAP表达增加,暴露于高脂饮食和/或追加缺氧可加重这种改变。 [Objective] To study the influence of intrauterine hypoxia on the expression of glial fibrillary acidic protein (GFAP) in the cerebral white matter of the elderly offspring of rats. [Methods] S-D rats intrauterine hypoxia model was estabhshed,then their offspring were exposed to high-fat diet (HF) and (or) second hypoxia (SH). Immunohistochemistry was used to detect the expression of GFAP in the white matter beside the lateral ventricle of the elderly offspring. Meanwhile the pathological image analysis system was used to determine the quantity. [Results] The main effect of intrauterine hypoxia increased the expression of GFAP in the white matter of the elderly offspring of rats (P 〈0.05). Interaction existed not only between IH and HF,IH and SH,or HF and SH (P 〈0.05) but also among IH, HF and SH (P 〈0.05). [Conclusion] Intrauterine hypoxia increases the expression of GFAP in the cerebral white matter of the elderly offspring of rats, which are aggravated by high-fat diet and(or) second hypoxia.
