摘要
为了实现对果蝇单个特定中枢神经元的分离,在传统急性机械分离方法的基础上,发明了一种使用微操纵器控制嵌有钨丝的玻璃电极,结合荧光成像,分离果蝇幼虫单个特定中枢神经元的新技术.在荧光共聚焦显微镜和倒置荧光显微镜下均观测到带有绿色荧光的果蝇幼虫Murashka神经元,并成功地分离出单个Murashka神经元,运用膜片钳技术记录到了Murashka神经元的钾电流.该技术可广泛应用于荧光标记的果蝇突变体中枢神经元的分离和相关研究.
Based on the traditional mechanical acute-isolation method, we developed a novel technique to isolate single specific drosophila larva CNS neuron by using two micro-manipulated glass pipettes with tungsten threads combined with fluorescence imaging. Under confocal and inversed fluorescence mieroseopies, we ob- served drosophila larva Murashka neurons, and successfully isolated one single Murashka neuron from drosophila larva. In the meantime, we recorded K current in this neuron using the patch-clamp technique. This novel technique is a powerful tool for isolation and investigation of fluorescence-labeled drosophila mutant CNS neurons.
