血管内皮细胞生长因子受体Flt-1与胃癌生物学行为的关系 被引量:1

Relationship between Expression of Flt-1 mRNA and Biological Behavior in Gastric Carcinoma

摘要 [目的]探讨血管内皮细胞生长因子受体fms-样酪氨酸激酶(Flt-1)在胃癌组织中的表达和胃癌组织中微血管密度(MVD)计数与胃癌临床病理因素及预后的关系。[方法]应用原位杂交和免疫组织化学技术,检测118例胃癌组织中Flt-1 mRNA及CD34蛋白的表达。[结果]胃癌组织中Flt-1 mRNA阳性表达率为55.9%;浸润性生长的肿瘤组织中Flt-1阳性表达率和MVD值明显高于膨胀性生长者(χ2=5.97,P<0.05,t=3.91,P<0.01),Flt-1 mRNA表达及MVD值与浸润深度(χ2=29.29,P<0.01,t=6.41,P<0.01),血管侵犯(χ2=27.70,P<0.01,t=7.96,P<0.01),淋巴结转移(χ2=28.40,P<0.01,t=8.45,P<0.01),肝转移(χ2=21.13,P<0.01,t=7.65,P<0.01)和腹膜转移(χ2=37.38,P<0.01,t=10.46,P<0.01)显著相关;MVD值与Flt-1 mRNA的表达水平有关,Flt-1 mRNA表达阴性和阳性患者的MVD值差异有显著性(t=11.24,P<0.01);Flt-1 mRNA阳性表达患者的平均生存时间和5年生存率均显著低于Flt-1 mRNA阴性表达患者(t=4.03,P<0.01,χ2=8.21,P<0.01)。MVD值≥54.9个/mm2患者的平均生存时间和5年生存率亦均低于MVD值<54.9/个mm2者(t=5.31,P<0.01,χ2=7.52,P<0.01)。[结论]Flt-1 mRNA阳性表达与胃癌血管生成有关,并参与肿瘤浸润转移的过程,可以作为反映胃癌生物学行为和判断预后的生物学指标及基因治疗的靶点。
