摘要 本研究目的在于调查一种抗肿瘤药Carboplatin(100m/kg)对灰鼠前庭器官功能与形态的影响。注射Carboplatin后,冷水诱发眼震持续时间明显缩短。急性形态学损害主要发生在Ⅰ型毛细胞的传入神经末梢和胞核。另外,壶腹嵴的形态学损害比球囊斑和椭园囊斑发生早而且更为严重。在囊斑内,微纹区是病变的主要部位。用药后30天,壶腹嵴和囊斑有较多Ⅰ型毛细胞缺失,然而Ⅱ型毛细胞未受明显影响。上述结果表明Carboplatin具有迅速的强烈的前庭毒性作用,Ⅰ型毛细胞和传入神经末梢是主要受损部位。 The purpose of this study was to examine the functional and morphological changes in the vestibular system of the chinilla after administering carboplatin(100mg/kg),an antineoplastic agent. The period of austained nystagmus evoked by cold caloric stimulation decreased significantly post-carboplatin treatment. Acute morphological damage was observed 1- 3 days post -treatment on the chalice of the afferent synapse beneath type Ⅰ hair cells and in the nuclei of type Ⅰ hair cells;type Ⅱ hair cells and their nerve endings appeared normal at this time. In addition,the morphological damage occurred earlier and was more severe in the crista than in the maculae of the saccule and utricle. Within the macula,the main locus of damage occurred in. the striola. At 30 days post-treatment,the majority of type Ⅰ hair cells were missing in both the crista and maculae;however,nearly all of the type Ⅱ hair cells were intact. These results indicate that carboplatin has extremely and rapidly vestibulotoxic effect that primarily targets the tyne Ⅰ hair cells and their afferent terminals.
