摘要 本研究探讨As2O3对肿瘤细胞中COX-2、MMP-2和MMP-9表达的影响。以体外培养SGC7901和K562细胞为对象,加不同浓度As2O3干预40和72小时。采用Western blot法和荧光定量RT-PCR法分别测定COX-2、MMP-2和MMP-9的表达和SGC7901细胞内的COX-2和MMP-2 mRNA的水平。结果显示:As2O3干预后SGC7901和K562细胞内的COX-2、MMP-2和MMP-9的表达水平呈时间和剂量依赖性减低;MMP-2 mRNA和COX-2 mRNA减低。结论:三氧化二砷可抑制肿瘤细胞COX-2、MMP-2和MMP-9表达,这一机制可能参与砷剂抑制肿瘤血管生成的过程。 This study was aimed to investigate on effect of As2O3 on expressions of COX-2, MMP-2 and MMP-9 in SGC7901 and K562 cells. SGC7901 and K562 cells were cultured in RPMI 1640 medium and were inoculated in culture medium with different concentrations of As2 O3 and at different times. Expressions of COX-2, MMP-2 and MMP-9 in SGC7901 and K562 cells were measured by using Western blot, while the levels of COX-2 mRNA and MMP-2 mRNA were measured with fluorescence quantitative RT-PCR. The results showed that the expression of COX-2, MMP-2 and MMP-9 decreased in dose- and time-dependent manners after treating with As2O3. The levels of COX-2 mRNA and MMP-2 mRNA reduced in groups treated with As2O3. In conclusion, As2O3 inhibits expressions of COX-2, MMP-2 and MMP-9 in K562 and SGC7901 cells, suggesting that As2O3 inhibits tumor development through its effect on angiogenesis involved in solid and hematologic malignancies.
