摘要
目的研究RGS4对常氧和低氧培养条件下HUEVC细胞凋亡的影响及地塞米松在其中的作用,以探讨RGS4在HUEVC细胞凋亡中的作用及地塞米松处理对其的影响。方法采用O2/N2/CO2三气培养箱模拟常氧和低氧条件培养HUEVC细胞,RGS4真核表达载体转染HUEVC细胞后采用流式细胞仪(Annexin-V-FITC-PI法)检测细胞凋亡。结果常氧条件下RGS4转染细胞和地塞米松处理对细胞凋亡无明显影响,低氧培养条件下细胞凋亡增加(P<0.01),RGS4转染后可使HUEVC细胞凋亡显著减少(P<0.01),地塞米松和RGS4同时处理后HUEVC细胞的凋亡与单纯RGS4转染无显著差别。结论RGS4不影响常氧条件下的HUEVC细胞凋亡,但可明显抑制低氧条件下的HUEVC细胞凋亡,地塞米松抑制细胞凋亡的作用可能与RGS4有关或者通过相同的细胞信号通路抑制低氧条件下的细胞凋亡。
Objective To study the role of RGS4 in apoptosis of HUEVCs in normoxia or hypoxia and the effect of dexamethasone on it. Methods HUEVCs were cultured in the simulated normoxia or hypoxia of O2/N2/CO2 cell incubator and were transfected with the eukaryotic expression vector of RGS4 ; the flow eytometer ( Annexin - V - FITC - PI) was applied to detect its apoptosis. Results The transfecfion of RGS4 and treatment of dexamethasone had no obvious effect on the apoptosis in normoxia while the apoptosis of HUEVCs increased in hypoxia (P 〈 0.01 ) ; the apoptosis of HUEVCs decreased significantly after RGS4 transfection ( P 〈 0.01 ) ; the apoptosis of HUEVCs treated with dexamethasone and with RGS4 at the same time showed no obvious difference from that treated with RGS4 transfection only. Conclusions RGS4 has no effect on the apoptosis of HUEVCs in normoxia but it can inhibit the apoptosis of HUEVCs in hypoxia; the effect of dexamethasonc on the apoptosis of HUEVCs is probably related to RGS4, or RGS4 and dexamethasone employ the same cell signaling pathway when inhibiting the apoptosis of HUEVCsw.
