摘要
目的运用骨生物力学、MicroCT分析及三维结构重建的方法,从“骨密度检测一形态性观察一功能学测试”角度出发,观察雄激素对去卵巢大鼠骨密度、骨微结构及骨力学性能的影响。方法30只6月龄雌性sD大鼠随机分为去势组(OVX)、治疗组(T)和假手术组(Sham),每组10只。治疗组大鼠去势后用睾丸酮水溶液灌胃给药,3个月后取材。取大鼠k椎体及右股骨行生物力学测试,取左股骨远端干骺部行MicroCT分析及三维结构重建。结果T组大鼠腰椎和股骨远端骨密度值(BMD)升高,骨小梁体积(BV/TV)、骨小梁厚度(Tb.Th)值升高,骨小梁间隙(Tb.Sp)值降低,而骨小梁数目(Tb.N)值无明显改变,腰椎各生物力学参数值升高,而股骨力学性能无明显提高,大鼠体重、子宫重量及股骨中点皮质骨厚度无明显的改变。结论雄激素可部分改善OVX大鼠骨小梁微观结构,防止骨量丢失,提高松质骨的力学性能,而对皮质骨和子宫无明显影响。
Objective To investigate the effect of androgen on bone in female ovariectomized rats(OVX) and reveal its mechanism, by using the Micro CT scanning and bone biomechanics test. Methods Thirty 6-month-old female SD rats were randomly divided into three groups including ovariectomized rats (OVX), sham-operated (Sham) and androgen (T) group, ten in every group. Femurs and vertebrae were then harvested 12 weeks after OVX for micro-computed tomography (Micro CT) scanning and biomechanical test. Results The administration of testosterone may reverse the decreasing BMD of OVX group, T group's BV/TV, Tb. Th enhanced while Th. Sp lower. Moreover, the testosterone significantly increased the ultimate stress and maximum load of lumbar vertebrae of the OVX rats. Conclusions These findings show that the testosterone has antiosteoporotic effects and can prevent the OVX-induced bone loss, mend bone microstructure and enhance cancellous bone intensity and BMD, but the testosterone effects slightly on the cortical bone, body weight and uterus.
