全酶法制备甘油二酯的研究(Ⅰ)——油脂酶法部分水解反应工艺条件的优化 被引量:3

摘要 通过对酶催化油脂部分水解反应和酶催化酯化反应的有效整合,提出了全酶法制备甘油二酯的工艺。主要研究了此工艺中的油脂(以菜籽油为例)部分水解反应,探讨了酶的种类、加水量、反应体系pH、反应温度、加酶量等因素对反应结果的影响,并对水解产物进行分子蒸馏提纯。较优的反应条件为:脂肪酶采用Lipozyme 20000L,加酶量为20 U/g,加水量为油重的10%,反应温度40℃,反应时间6 h。油脂部分水解反应产物经过分子蒸馏提纯后,可以获得含量在50%以上的甘油二酯产品。 The holoenzymatic process combining the enzymatic partial hydrolysis with enzymatic esterification was developed to produce diacylglycerol. Optimization of enzymatic partial hydrolysis of rapeseed oil,the part of the holoenzymatic process,was studied. The influence of factors on reaction, including the type of lipase, pH of reaction system, initial water content, enzyme dosage, were decided, and the product of hydrolysis reaction was further purified by molecular distillation. Optimal conditions were determined as follows :Lipozyme 20000L lipase, enzyme dosage 20 U/g, initial water content 10% (based on the weight of oil), reaction temperature 40℃, reaction time 6 h. After molecular distillation, the content of diacylglycerol in finished product was over 50%.
