摘要
研究了青霉Q59对棉秆的降解作用。测定了菌株的产酶动力学,通过正交试验检测了棉秆粒径、发酵液pH、发酵温度及发酵时间4个因子对棉秆糖化率的影响。结果表明,该菌株CMC酶活在第9天达到高峰;以10%麸皮汁作为氮源,Q59的CMC酶活、FPA酶活及糖化率均为最高;正交试验确定了Q59发酵棉秆的优化工艺参数,即将棉秆粉碎至10目,维持发酵液在pH5~6范围,于37℃发酵6~9d,能获得较高的糖化率。
The degradation of cotton stalk by penicillium Q59 was studied. Cellulase activity dynamics was determined firstly, and orthogonal test was designed to investigate the effects of cotton stalk grain size, pH value of fermenting liquid, fermentation temperature and fermentation time on saccharification rate of cotton stalk. The results showed that the highest CMCase activity presented on the ninth day and the highest CMCase activity and FPase activity and sacchrification rate of Q59 presented as 10 % bran juice used as nitrogen source. The optimum fermentation technical parameters of Q59 to achieve high saccharification rate were determined by orthogonal test as follows: cotton stalk grinded in 10 mesh, pH value of fermenting liquid maintained within 5- 6, 6- 9 d fermentation at 37℃.
