摘要
以无菌生根苗的叶片和茎段为外植体进行了尾赤桉无性系DH201-2不定芽的诱导及植株再生。通过对外植体的筛选、激素种类和浓度的调整、培养条件的选择,结果表明,取自生根培养基上培养30 d植株的上部和中部的茎段在改良MS+TDZ 0.05 mg.L-1+NAA 0.50 mg.L-1的培养基中获得高达57.0%的丛生芽诱导;取在生根培养基上培养20 d的植株,顶端叶片在改良MS+TDZ 0.05 mg.L-1+IBA 0.10 mg.L-1的培养基中获得48.3%的植株再生;适合于茎段和叶片不定芽分化的Ca(NO3)2浓度为0.706 0 g.L-1。
In vitro seedling leaves and stems were used as explants to study the shoot inducing and plant regeneration of Eucalyptus urophylla × E. camalduleusis clone DH201-2. The results indicated that: the leaf explants should be taken from the upper section and the stem explants should be the middle and upper section; The optimal medium of regeneration were MS + TDZ 0.05 mg · L^-1 + IBA 0.10 mg · L^-1 for leaf explants and MS + TDZ 0.05 mg· L^-1 + NAA 0.05 mg · L^-1 for stem explants, with regeneration rates of 48.3% and 57.0% respectively; The proper concentration of calcium nitrate were 0.706 0 g · L^-1 for the induction of adventitious shoots from leaves and stems explants ; Leaf explants from plantlets with a rooting culture of 15 days had a high regeneration rate, while plantlets with a rooting culture of 30 days were the best for the collection of stem explants. Most of the explants can produce clump shoots.
