白藜芦醇减轻内皮细胞损伤与活性氧的表达 被引量:1

Resveratrol Attenuated Reactive Oxygen Species in Injured Endothelial Cells

摘要 背景白藜芦醇是红葡萄酒中具有心血管保护作用的最重要的物质,可通过其强大的抗炎作用发挥抗动脉粥样硬化作用。目的观察白藜芦醇对因肿瘤坏死因子α(TNF-α)或sCD40L损伤的人脐静脉内皮细胞(HUVEC)的增殖活力和活性氧簇(ROS)的影响。方法体外培养HUVEC,白藜芦醇(1~50μmol/L)预孵育2h后,加入TNF-α(10μg/L)或sCD40L(10μg/L)刺激4h。用MMT法检测各组内皮细胞的活力;利用免疫荧光法检测各组HUVEC内活性氧的表达。结果TNF-α和sCD40L均可损伤HUVEC,使细胞增殖活力明显下降(分别为对照组的32.7%和26.0%,P<0.05),ROS表达增加(分别为对照组的3.4倍和4.9倍,P<0.05),而白藜芦醇(10、50μmol/L)可有效预防TNF-α和sCD40L对HUVEC的损伤(P<0.01)和抑制HUVEC内ROS的产生(P<0.01)。结论白藜芦醇可通过抑制HUVEC内ROS的产生,降低TNF-α或sCD40L对细胞的氧化损伤,发挥保护血管内皮的作用。 Background Resveratrol has been unanimously recognized as an cardiovascular protective substance in red wine. It has been speculated that the anti-atherosclerosis effect of resveratrol is ascribed to its powerful anti-inflammatory effect. Objective To investigate the effects of resveratrol on injured human umbilical venoendothelial ceils (HUVEC) and the reactive oxygen species(ROS) production induced by TNF-α or soluble CD40L (sCD40L). Methods Cultured HUVEC were pre-incubated with resveratrol(1- 50 μmol/L) for 2 hours and then treated with TNF-α( 10μg/L) or sCD40Lα( 10 μg/L) for another 4 hours. MTT assay was used to detect proliterative activity of HUVEC. Immunofluorescence microscopy was used for determination of ROS expression. Results Both TNF-α and sCD40L impaired HUVEC proliferation (-32.7% and -26% vs control, P〈0.05) which was associated with increases in ROS expression by 3.4 and 4.9 folds respectively (P〈0.05). Resveratrol (10 μmol/L, 50 μmol/L) dose-dependently prevented the cells from being injured by TNF-α and sCD40L (P〈0.01), and decreased the expression of ROS(P〈0.01 ). Conclusion Resveratrol protect TNF-α or sCD40L-induced injury in HUVEC might be ascribed to inhibition of generation of ROS.
