利用GenBank中获得的近缘物种微卫星序列设计的20对引物,对河鲈养殖群体20个个体基因组DNA进行扩增,发现8对引物能扩增出特异性谱带,获得32个等位基因,5个位点的等位基因数大于或等于4个,多态信息含量0.3680~0.7395,5个位点多态信息含量为高度多态(PIC>0.5)。期望杂合度范围在0.4154~0.7936,观测杂合度普遍高于期望杂合度。个体识别率范围在0.180~0.620,累积个体识别率为0.9899,单一微卫星位点的个体识别率普遍低于0.8,未出现高度多态性遗传标记。非父排除率范围在0.2537~0.6185,累积非父排除率0.9937,4个位点的非父排除率高于0.5,属于高度多态性遗传标记。
The total DNA isolated from 20 individuals of cultivated population of Perca fluviatilis was amplified with 20 primers which were designed according to microsatelhte sequences of closer species from GenBank database. Only eight of twenty primers gave specific fragment pattern, and 32 alleles were obtained. There are five loci whose Alleles number was not less than four. The polymorphism information content (PIC) of each loci was from 0.3680 to 0. 7395, five of which were high polymorphic ( PIC 〉 0.5 ). The unbiased expected heterozygosity was in the range of 0.3000 to 1. 01300, and the number of observed heterozygosity was in the range of 0.4154 to 0.7936, respectively, so the number of observed heterozygosity was greater than the unbiased expected heterozygosity. The power of discrimination (DP) was in the range of 0. 180 to 0. 620, and the cumulate power of discrimination was 0. 9899, which showed that high polymorphie genetic marker doesn't occur because of the power of discrimination ( DP 〈0.8). The probability of paternity exclusion was in the range of 0.2537 to 0.6185, and the cumulate probability of paternity exclusion was 0.9937. Only four of eight loci whose the probability of paternity exclusion was greater than 0.5 were high polymorphic.
