摘要
本研究探讨了生长激素(STH)和胰岛素对猪卵母细胞体外成熟及孤雌激活后卵裂的影响。结果表明:在mNCSU-23液中,添加0.15μg/mLSTH组的成熟率(73.83%)和卵裂率(64.76%)显著高于对照组和添加0.01、0.05、0.1、0.2μg/mLSTH组(P<0.05);添加5μg/mL和8μg/mL胰岛素组的成熟率(66.25%、60.64%)显著高于添加0、0.5、2μg/mL及10μg/mL组(P<0.05);添加5μg/mL胰岛素组的卵裂率(61.63%)显著高于添加0、0.5、2、8、10μg/mL组(P<0.05);最佳的联合添加组为0.15μg/mLSTH+2μg/mL胰岛素,其体外成熟率为79.37%、卵裂率为72.31%。
This experiment investigated the effects of STH and insulin on porcine oocytes maturation in vitro and cleavage after parthenogenetic activation. The results showed that the maturation rate (73.83%) and the cleavage rate (64.76%) derived from oocytes matured in mNCSU-23 supplemented with 0.15 ug/mL STH were significantly higher than those supplemented with 0.01, 0.05, 0.1, 0.2 ug/mL and control group (P 〈 0.05). When 5 ug/mL or 8 ug/mL insulin was added to mNCSU-23, respectively, the maturation rate (66.25%, 60.64%) were significantly higher than that of other groups(P 〈 0.05), and the cleavage rate(61.63%) matured in mNCSU-23 supplemented with 5 ug/mL insulin was significantly higher than that of other groups (P 〈 0.05). The optimum concentrations for STH and insulin to use together were 0.15 ug/mL and 2 ug/mL, the rate of maturation and cleavage were 79.37%, 72.31%, respectively.
