雌激素对兔腹主动脉球囊损伤反应的影响

摘要 目的探讨雌激素对兔腹主动脉球囊损伤反应的影响。方法日本雌性和雄性大耳白兔60只,随机分为假手术组、对照组、干预组,每组雌、雄各10只。假手术组行股动脉切开术,另两组以球囊导管损伤兔腹主动脉内膜,制作球囊损伤模型,干预组每天肌注17-β雌二醇(17-βE_2、100μg/kg),另两组每天肌注300μl棉子油。2周后,测定NO和内皮素(ET-1)后;动物处死取腹主动脉标本,作病理学切片,HE染色,测定内膜面积、中膜面积、内膜面积/中膜面积比值;免疫组织化学染色增殖细胞核抗原,并计算其增殖指数(PI)。结果与假手术组比较,干预组雄性兔和对照组雄、雌性免血管壁增厚、管腔变小,NO含量下降(P<0.01),ET-1含量上升(P<0.01),干预组雌性兔变化不显著(P>0.05)。干预组雄性兔和对照组雄、雌性兔ET-1、内膜面积、中膜面积、内膜面积/中膜面积比值、PI均较假手术组增加(P<0.01),而干预组雌性兔ET-1较假手术组无显著变化(P>0.05)。内膜面积、中膜面积、内膜面积/中膜面积比值、PI虽较假手术组增加(P<0.01),但较对照组明显减少(P<0.01)。结论雌激素对雄性兔血管球囊损伤无干预作用,对雌性兔血管球囊损伤有保护作用。 Objective To investigate effect of estrogen on balloon-injury reaction of rabbit abdominal aorta. Methods Sixty rabbits were randomly divided into 3 groups(10 males and 10 females each) :sham-operation group, control group and estradiol therapy group. The rabbit abdominal aorta balloon injury model was established in control group and estradiol therapy group. Sham-operation group and control group received daily injection of 300/11 cottonseed oil. Estradiol therapy group received daily injection of estrogen(17-β-estradiol 100 μg/kg). Two weeks later, the blood samples were used to assay nitric oxide(NO) and endothelin(ET-1). The abdominal aortas were taken for pathologic analysis and proliferating cell nuclear antigen(PCNA) staining. The proliferation index(H) was calculated. Results Abdominal aorta wall was thicker, lumen was smaller,NO was decreased and ET-1 was increased in control group and male estradiol therapy group than in sham-operation group (P〈0.01), but those were not changed in female estradiol therapy group compared with sham-operation group (P〉0.05). ET-1 ,PI,intima and media area, ratio of intima/media (I/M) were increased in control group and male estradiol therapy group compared with those in sham-operation group (P 〈 0.01). ET-1 was not changed in female estradiol therapy group compared with sham-operation group (P〉0.05). PCNA,PI, intima and media area,ratio of I/M were increased in female estradiol therapy group compared with sham-operation group (P 〈 0.01),but these were significantly decreased in female estradiol therapy group com- pared with control group (P 〈 0.01). Conclusion Estrogen has protective effect against balloon injury of female rabbit abdominal aorta,but not on male rabbit.
