高氧液对家兔心肌缺血再灌注损伤的保护作用与Hsp70的关系

摘要 目的研究高氧液对家兔心肌缺血再灌注损伤的保护作用与热休克蛋白Hsp70的关系。方法56只家兔随机分为3组。动物接受冠状动脉左前分支60min阻断和120min再灌注。分别取心肌检测不同时点Hsp70的表达;检测超氧化物歧化酶活性、丙二醛含量;测量危险区心肌梗死面积。结果HBO组、IR组SOD活性均较P组降低;MDA含量则较P组升高。心肌梗死面积HBO组较IR组比较差异有显著性,分别为(25.2±4.2)%、(45.5±4.3)%。Hsp70的表达HBO组较IR组上调。结论高氧液处理可降低心肌梗死面积;保护SOD、抑制MDA生成,上调Hsp70的表达。 Objective To investigate the effects of hyperbaric oxygen and the relationship with Hsp70 on myocardial ischemia and reperfusion injury of rabbits. Methods 56 family rabbits were allocated to each of 3 groups(n = 8) randomly:pseudo-operation group(group P),ischemia and reperfusion group(group IR),hyperbaric oxygen group(group HBO). Animals in each group except group P, experienced LDA occlusion for 60 minutes and reperfusion for 120 minutes. Animals in group HBO received hyperbaric oxygen infusion before ischemia and during reperfusion. Activity of superoxide dismutase( SOD ), quantity of malondialdyde ( MDA ) in myoeardium was detected, and infarct size was caculated. Results MDA quantity in group IR, HBO raised significantly and activity of SOD decreased significantly compared with group P. Infarct size of area at risk in group HBO(25.2 ± 4.2)% was significantly lower than that in group IR(45.5 ± 4.3 )%. The expression of Hsp70 was significantly up-regulated in group HBO during reperfusion. Conclusion Hyperbaric oxygen infusion could reduce infarct size of area at risk, protect the activity of SOD and reduce the product of MDA, up-regulate the expression of Hsp70.
