摘要
目的研究氯化甲基汞(MMC)对急性早幼粒细胞性白血病(APL)模型裸鼠的作用。方法去除免疫力BALB/C裸鼠经3Gy^(60)Co照射10min,尾静脉接种5×107/ml对数生长期NB4细胞0.1ml,建立APL裸鼠模型。隔2日灌胃给予10mg/kg(0.1ml/10g)的MMC或三氧化二砷(ATO)(ATO作抗癌阳性对照),探讨MMC对APL裸鼠白细胞计数及分类、脾/体比值及生存期等的影响。结果MMC能降低APL裸鼠的白细胞计数、白血病细胞比例及脾/体比值,延长APL裸鼠生存期,且与APL对照组比较,差异有统计学意义(P<0.05)。结论4MMC对APL裸鼠各项指标有一定影响,可通过进一步的机制研究探讨将其用于治疗。
[ Objective] To study the effect of methyl mercury chloride (MMC) on nude mice of APL model. [ Methods] 0.1 ml ( 5 ×10^7/ml) NtM cells were given to the BALB/C nude mice by tail vein inoculation after irradiation of 3 Gy 60Co for 10 minutes to make APL mice model. APL mice were administrated orally with 10 mg/kg(0.1 ml/10 g) BW MMC or AT0(ATO as positive control for anti-cancer) once after two days. The effects of MMC on nude mice's leukocyte count and classification, spleen weight/body ratio and the survival time were observed. [ Results] MMC could reduce nude mice's leukocyte count and spleen weight/body ratio, extent the survival time, comparing with control group, the difference was significant ( P 〈0.05). [ Conclusion] MMC has a certain influence on indicators of APL nude mice, through further research, its mechanism can be explored for the treatment.
