软脂酸对阿霉素诱导的原代MEF细胞DNA损伤反应的调节

摘要 目的探讨软脂酸是否影响原代MEF细胞的DNA损伤反应。方法利用western方法检测原代MEF细胞p53及p-p53蛋白的表达变化,判断软脂酸是否影响原代MEF细胞的DNA损伤反应。结果(1)软脂酸抑制阿霉素诱导的原代MEF细胞p53蛋白的表达。(2)软脂酸抑制阿霉素诱导的原代MEF细胞p53蛋白的磷酸化。结论软脂酸可能通过抑制DNA损伤反应,有利于损伤细胞转化为肿瘤细胞。 Objective To detect whether palmitic acid can regulate DNA damage response in primary MEFs. Methods We studied the role of Palmitic acid in DNA damage response in primary MEFs by checking p53 acctunulafion by western blot analysis. Results (1) Palmific acid inhibited adriamycin-induced expression of p53 in primary mouse embryonic fibroblasts. ( 2 ) Palmitic acid inhibited adriamycin-induced activation of p53 in primary mouse embryonic fibroblasts. Conclusion These results suggest that Palmitic acid,by inhibiting DNA damage response,might promote cell transformation into turnout cell.
