SLE中淋巴毒素β基因表达及相关性研究

摘要 目的:探讨LT-β在SLE发病及免疫调节紊乱中的作用。方法:用ELISA法检测血清LT-β水平,PBLRNA斑点杂交测定LT-β基因转录水平;PCR-RLFP法分析LT-β基因多态性。结果:SLE患者血清LT-β水平比正常显著增高(P<0.05);活动期比正常增高更显著(P<0.01)。SLE患者基因转录水平比正常对照显著增高(P<0.05);活动期比正常亦显著增高(P<0.05);未发现LT-β基因的XhoⅠ酶切位点具有多态性。结论:提示LT-β是导致SLE发生的主要因子;LT-β参与SLE的免疫调节紊乱。 Objective:To study expression of lymphotoxin-β(LT-β) in SLE patients with the relationship of inordinate immunoregulation. Methods: The 82 SLE patients and 90 normal controls' serum concentration of LT-β protein was detected with ELISA and PBLs were sepa- rated for analyses of its encoding sequence with RNA spot hybridization. The polymorphisms of LT-13 gene in PBLs were analyzed by PCRRFLP. Results:Tbe expressing level of LT-β in SLE patients was significantly higher than in controls for both seral protein and mRNA inPBLs (P 〈 0.05) ;where those in active group was much higher (P 〈 0.01 ). Hybridization by using LT-β probe was showed significant difference between SLE patients and control groups. ( P 〈 0.05 ) ;and it was significantly higher in active group than in controls( P 〈 0.05). There was no polymerism variation found in Xho I site of LT-β gene in both groups. Conclusion: The results suggest that LT-β is the main cytokine related with SLE which is involved in the inordinate immunoreglution in SLE.
