摘要 目的了解不同剂量的泼尼松制剂对小鼠特异性和非特异性免疫功能的影响。方法以肌肉注射0.25 mL/kg、0.124 8 mL/kg、0.05 mL/kg的用药量将小鼠分为高、中、低剂量组,同时设一对照组,连续用药10 d,分别检测各组小鼠的体质量、胸腺指数、脾脏指数、巨噬细胞的吞噬率、中性粒细胞的吞噬率、T淋巴细胞E花环率、溶血素效价和抗绵羊红细胞抗体形成细胞的水平。结果高剂量组与对照组相比,除巨噬细胞的吞噬率、T淋巴细胞E花环率、胸腺指数无显著性差异外,其余各项指标都有显著性差异;中剂量组、低剂量组与对照组相比,体质量、胸腺指数、脾脏指数、中性粒细胞吞噬率、溶血素效价有显著性差异,其余各项指标无显著性差异。结论高剂量的泼尼松制剂对体质量、脾脏质量、中性粒细胞吞噬功能、B淋巴细胞分泌抗体的能力、抗绵羊红细胞抗体形成细胞的水平有抑制作用。中、低剂量的泼尼松对体质量、脾脏质量、胸腺质量、中性粒细胞吞噬功能、B淋巴细胞分泌抗体的能力有抑制作用。 Objective It is to explore the effects of prednisone of different dosage on specific and non-specific immune function of rats. Methods Rats were divided into high dosage, mild dosage and low dosage group according to the used dosage of 0.25 mL/kg, 0. 124 8 mL/kg and 0.05 mL/L by intramscular injection. Another group of rats were chosen as control group. After 10 days' treatment, the index of thymus gland and spleen, function of abdominal cavity macrophage and neutrophil, antibody production(SRBC- PFC) and serum haemolysin titers, E- rosette formation were observed respectively. Results There were significant differences in all the indexes except for phagocytosis rate of phagocyte, SRBC- PFC and index of thymus gland (P 〈 0.05 ) in the high dose group compared with the control group. There were no significant differences in the indexes except for thymus index, spleen index, neutrophil function, production of the hemolysin antibody in the middle dose group and low dose group compared with the control group. Conclusion High dose of prednisone has inhibition on body weight, spleen weight, phagocytosis of phagocytes, antibody production of B lymphocytes and E- rosette formation. Mild and low dose of prednisone has inhibition on body weight, spleen weight, thymus gland weight, phagocytosis of phagocytes, antibody production of B lymphocytes.
