组织型纤溶酶原激活因子及其抑制因子在孕鼠子宫中的表达 被引量:1

摘要 本实验用原位杂交和免疫组织化学定位方法从mRNA和蛋白质水平研究了组织型纤溶酶原激活因子(tPA)及其I型抑制因子(PAI-1)在受孕第七天大鼠子宫中的表达。结果表明,在非着床点子宫腔上皮细胞、腔上皮附近的基质细胞和腺上皮细胞中,tPA和PAI-lmRNA都有表达,但PAI-1mRNA的量很低。tPA抗原分布于子宫腔上皮和腺上皮,PAI-1抗原只能在腺上皮和表层基质中微弱地检测到。在着床点,tPA和PAI-1的mRNA的抗原大量分布于蜕膜。此结果提示,tPA可能参与子宫内膜的蜕膜化等组织改组过程;PAI-1可能在蜕膜中发挥保护作用,阻止胚泡过度侵入。 Using in situ hybridization and immunohistochemical methods ,we studied the expression of both tissue type plasminogen activator (tPA ) and plasminogen activator inhibitor type-l (PAI-1 ) mRNA and antigen in the implantation sites and nonimplantation segments of preg- nant rats on Day 7. The results showed that ,tPA and PAl-I mRNA localized in luminal ep- ithelial cells ,an underlying band of stromal cells ,and glandular epithelial cells in non-implan- tation segments. The quantity of PAI-1 mRNA was low. At the same time ,tPA antigen dis- tributed in luminal and glandular epithelium , while PAI- 1 antigen could only be weakly de- tected in glandular epithelium and superficial layer of stroma. Both tPA and PAI-1 mRNA in implantation sites were strongly expressed , mainly in decidual cells ,and their antigens were intersely present in the decidua. We suggest that the expression of tPA in the implantation sites may involve in the decidualization of endometrium and PAI-1 may play an essential role in protecting the uterus from excessive intrusion by trophoblasts.
