干细胞标志物Nanog的检测在胃癌诊断中的意义 被引量:29

摘要 目的检测胃癌患者癌组织和癌旁组织中干细胞标志物Nanog的表达水平并探讨其与临床病理参数的相关性。方法用RT-PCR和Real-timePCR检测62例胃癌患者癌组织和癌旁组织中Nanog的表达。结果RT-PCR结果显示胃癌组织和癌旁组织中Nanog的表达阳性率分别为58.1%(36/62)、9.7%(6/62),差异有统计学意义(P<0.0001);Real-timePCR结果表明胃癌组织中Nanog相对表达水平高于癌旁组织(P<0.05);胃癌组织中Nanog阳性表达与肿瘤分化状态相关,低、未分化组高于中、高分化组(P<0.05),但与年龄、性别、肿瘤大小、浸润深度、TNM分期和有无淋巴结转移无关。结论Nanog表达与胃癌的发生及其分化状态相关,可作为胃癌诊断的一个新的分子标志。 Objective To detect the expression levels of Nanog, a stem cell marker, in paired tumor and non-cancerous tissues of gastric cancer (GC) , and explore the correlation between Nanog expression and the clinicopathological parameters of GC. Methods The ex- pressions of Nanog in primary tumor tissue and their matching adjacent non-cancerous tissue from 62 GC patients were examined by re- verse transcription-PCR (RT-PCR) and real-time PCR. Results RT-PCR results showed that the positive rate of Nanog expression in gastric tumor tissues was 58.1%, which was significantly higher than that in the matching adjacent non-cancerous tissues (9.7% ,P 〈 0.0001 ). The relative level of Nanog expression in tumor tissues was also significantly higher than that in the paired adjacent non-can- cerous tissues (P 〈 0.05 ). The Nanog expression correlated with the differentiation state of GC. The positive rates of Nanog expression in the poorly differentiated and undifferentiated GC tissue were significantly higher than those in the moderately and well-differentiated GC tissues ( P 〈 0.05 ) , but no correlation was found between Nanog expression and other clinicopathological parameters, e.g. , age, gender, tumor size,TNM stage, depth of invasion, and lymph node metastasis of the GC patients. Conclusions The genesis of GC correlated with cancer stem cells (CSCs). Nanog expression was associated with the differentiation state of GC. Nanog may serve as a novel marker for diagnosis of human GC.
