摘要
目的探讨衰变加速因子(DAF)在宫颈癌中的表达及其生物学意义。方法以宫颈癌细胞系ME180、宫颈癌组织和癌旁正常组织为材料,采用real-time PCR和western blotting检测DAF基因的mRNA和蛋白质表达水平;用3H-胸腺嘧啶核苷酸掺入量(3H-thymidine incorporation,3H-TdR)检查宫颈癌细胞的增殖,用Transwell小室法评估宫颈癌细胞的迁移能力。结果DAF的表达在宫颈癌组织中明显高于其配对的癌旁正常组织,抑制DAF基因的表达能明显降低ME180细胞的增殖、迁移能力。结论DAF在宫颈癌细胞中的高表达可能是肿瘤细胞免疫逃逸的重要机制,对宫颈癌的发生发展有重要的生物学意义。
Objective To investigate the effect of decay-accelerating factor (DAF) on the survival of human cervical ovarian cancer cells. Methods The expression of DAF mRNA and protein was examined by real-time PCR and Western blot respectively. The effect of DAF on the migration of ME180 cell was assessed by Transwell and the cell proliferation was measured by 3H-thymidine incorporation. Results The expression of DAF was significantly increased in human cervical cancer tissue, and the suppression of DAF expression con- siderably decreased the migration and proliferation of ME180 cells. Conclusions The high expression of DAF may involve in the immune escape of cancerous cells, so its biological effects are important for carcinogenesis and development of human cervical ovarian cancer.
