摘要
目的:观察足叶乙甙(VP16)对人粘液表皮样癌高转移细胞株(Mc3)抑制作用。方法:采用MTT及软琼脂克隆形成法观察VP16对Mc3的药敏性。结果:VP16对Mc3细胞的生长及克隆形成有明显的抑制作用,且呈显著的剂量依赖性。MTT药物敏感性试验VP16对Mc3细胞作用IC50值为1412μg/L,RAA值为5.9;克隆形成法IC50值为126μg/L,RAA值为93.4。结论:Mc3细胞对VP16有较高的敏感性。VP16在人粘液表皮样癌的治疗,特别是肿瘤转移的防治方面可能有一定意义。
Objective: To study the inhibitory effects of Etoposide(VP16)on a human highly metastatic mucoepidermoid carcinnoma cells (Mc3). Methods: Mc3 cells were exposed to VP16 at various concentrations. The inhibitory effects of VP16 were tested with microculture tetrazolium colorimetric assay(MTT)and colonogenic assay respectively. Results: VP16 inhibited in a dose dependant manner in both assays.The IC 50 and RAA values obtained in MTT assay were 1412 μg/L and 5.9 respectively, that is clonogenic assay were 126 μg/L and 93.4 respectively.Conclusion:VP16 may inhibit Mc3 cell growth and may be potential in the inhibition of Mc3 cell metastasis.
