摘要
以香花槐的新鲜花粉为试验材料,利用TTC(氯化三苯四氮唑)和碘-碘化钾染色法及培养基萌发试验,选配出最适合花粉萌发的培养基,以研究不同浓度的Ca2+对花粉萌发和花粉管伸长的影响。结果表明:香花槐花粉管萌发和花粉管生长的最适培养基是10mg/L蔗糖+2.0mg/L硼酸+0.8g琼脂,较低浓度的钙对花粉萌发和花粉管生长有促进作用,而较高浓度的钙对其有一定的抑制作用。
The fresh pollens of Robinia pseudoacacia CV. Idaho. were cultured on the optimum medium with different Ca2+ concentration to investigate the effect on its germination and pollen tube elongation. The results show that the optimum medium for pollen germination and pollen tube elongation of Robinia pseudoacacia CV. Idaho is 10 mg/L cane sugar+2.0 mg/ L boric acid+0.8 g agar, and the lower Ca2+ concentration promotes its pollen germination and pollen tube growth and high Ca2+ concentration inhibits its pollen germination and pollen tube growth.
