摘要
建立了高效液相色谱-蒸发光散射检测法(HPLC-ELSD)同时测定阿莫西林和6-APA(6-氨基青霉烷酸)中糖的新方法。乙腈-水为流动相,流速1.0 mL/min,柱温25℃,蒸发光散射检测器漂移管温度85℃,氮气作载气,流速2.00 L/min。糖的线性范围:果糖、葡萄糖、蔗糖、麦芽糖0.10-3.00μg,相关系数为:0.9991-0.9998,4种糖的加标回收率范围为88.30%-112.74%,检出限(S/N=3)低于7.00μg/mL。
A new approach for simultaneously determination of Carbohydrates in Amoxicillin and 6-Aminopenicillanic acid by High Performance Liquid Chromatography with Evaporative Light Scattering Detector (HPLC-ELSD) was developed. The carbohydrates were separated using an acetonitrile-water gradient system at a flow-rate of 1.0 mL/min. The optimum setting of the EISD parameters: drift tube temperature was 85 ℃ and nitrogen carrier gas flow-rate was 2.00 mL/min. The ab of the Carbohydrates (fructose, glucose, sucrose, maltose )showed distinct linearity relation ship with the concentration of the Carbohydrates in the range of 0.1 - 3.0 μg, the correlation coefficient was 0. 999 1 - 0. 999 8. Recovery of 88.3% - 112.7% was obtained, and the detection limit (SIN = 3)was found to be under 7 μg/mL.
