摘要
为增强抗白血病药物敏感性 ,采用液体培养法研究环孢霉素 A(CSA)与硫氮酮 (DZ)单用或合用分别增强 K56 2细胞、耐药 K56 2细胞 (K56 2 /Adr)对阿霉素 (Adr)、足叶乙甙 (VP16 )两种化疗药物敏感性的增强作用。结果表明在 K56 2及 K56 2 /Adr细胞中 ,单用硫氮酮较高浓度 (2 2 1.7μmol/L)时才呈现对 Adr或 VP16敏感性的增强作用 ;单用 CSA较高浓度 (4.2 μmol/L)时增强 Adr对 K56 2细胞的杀伤性而对 VP16增强作用则不明显 ,较高浓度 CSA(4.2μmol/L )均能提高 K56 2 /Adr细胞对Adr或 VP16的敏感性 ;采用低浓度的 CSA(1.3μmol/L )和硫氮酮 (2 .2μmol/L )联合作用于 K56 2 /Adr细胞 ,呈现对 VP16或 Adr药物敏感性的明显增加。提示 CSA与
In order to improve the effect of anti leukdmia drugs, cyclosporin A(CsA) and diltiazem were used in vitro singly and combinedly to study the increased sensitivity of sensitive cell strain K562 and resistant cell strain K562(K562/Adr) to adriamycin (Adr) and etoposide (VP16) in liquid culture . Sensitivity of K562 and K562/Adr to Adr or VP16 is only increased when exposed to higher concertration (221 7μmol/L) at the single application of diltiazem. When exposed to higher conertration (4 2μmol/L) of single CsA, increased sensitivity was showed in K562 to Adr and in K562/Adr to either Adr or VP16. However , when CsA and diltiazem were combinedly used at lower concertration (1 3μmol/L and 2 2μmol/L, respectively), sensitivity to Adr and VP16 was profoundly enhanced. It indicated that higher dose of CsA and diltiazem may improve the effect of chemotherpeutic drugs. Furthermore, combination of CsA and diltiazem, although at lower concertrations, will perform a cooperative function.
