摘要
目的:促进内皮细胞在聚氨酯材料上粘附,为实现人工心血管内皮细胞化摸索条件和方法。方法:用荧光法观察细胞外基质成分对内皮细胞在聚氨酯材料上粘附的影响。结果:用胶原Ⅰ、胶原Ⅳ、纤维粘连蛋白(FN)、玻璃粘连蛋白(VN)等细胞外基质成分及对照组牛血清白蛋白(BSA)分别预衬聚氨酯材料,在50mg/L浓度组,内皮细胞与材料接触15min后,粘附率分别为32.31%、54.43%、42.34%、63.77%和27.73%。与对照组相比,几种细胞外基质成分对内皮细胞都有明显的促粘附作用(P<0.01和P<0.05)。其中,VN的促粘附作用最好,胶原Ⅳ和FN的促粘附作用也较好;而胶原Ⅰ的促粘附作用较弱。结论:使用VN或联合使用VN和其他细胞外基质成分预衬聚氨酯材料。
Objective:To promote the adhsion of endothelial cells onto polyurethane (PU) material for realization of endothelialization on artificial heart and blood vessel.Methods:The influence of elements in extracellular matrix on the adhesive function of endothelial cells onto the PU material was observed with fluorescent method.Results:It revealed that collagen I,collagen Ⅳ,FN,VN precoated on the PU material could significantly enhance the adherance of endothelial cell,among which VN seemed to be the best.In concentration of 50 mg/L the cell attachment rate in collagen I,collagen Ⅳ,FN,VN group after 15 minutes contact were 32.31%,63.77%,54.43%,42.34%(P<0.05) and 27.73%(P<0.01),respectively.Conclusions:It suggests that precoating of PU material with VN or with VN and other ECM elements in combination could be lead to endothelialization.
