景天属药用植物遗传多样性的RAPD分析 被引量:10

摘要 采用RAPD技术对景天科景天属(Sedum)的凹叶景天、江南景天、宽叶景天、佛甲草和垂盆草等6种药用植物进行分子水平鉴定,并初步分析其相互间的亲缘关系。CTAB法提取各药用植物总DNA,以45条随机引物进行随机扩增,筛选出10条随机引物很好地区分了11个试样,扩增出208个位点,其中多态性位点173个(83.2%)。用Popgen32计算出各种间的Nei’s(1978)遗传距离和遗传相似性矩阵,采用UPGMA绘出了亲缘关系聚类图。结果表明RAPD法能有效地鉴别景天属6种药用植物,探讨其种间亲缘关系和种内遗传多样性,其结果还为同属待定种鉴定提供了DNA分子水平依据。 To identify the medicinal plants of Sedum L. , including Sedum emarginatum Migo. , S. kiangnanense D. Q. WAN, S. ellacombianum Praeger. , S. lineare Thunb. and S. sarmentosum Bunge. , by RAPD in molecular level,and preliminarily analyze the relationship among those species. Isolating general DNA from fresh leaves by CTAB and 45 random primers were used to identify them. 10 random prim- ers amplified 208 loci which have 173 polymorphic loci, namely 83.2%. The matrix of genetic distance and genetic identity in species was calculated through using Popgen32 by Nei' s(1978). Dendrogram was constructed by using UPGMA. RAPD is an effective method to identify the medicinal plants of Sedum L. , and can explore the relationship among species and genetic diversity within the species. The results also provide a basis of DNA molecular level for the identification of the species to be authenticated.
